Inhibition of aggregation of the slime mould Dictyostelium discoideum by a factor diffusing from Escherichia coli.

چکیده

A factor diffuses from Escherichia colt that temporarily or permanently prevents separate pre-aggregation cells of Dictyostelium discoideum from forming aggregation centres or streams. Centres are inhibited at a greater distance from the bacteria than are streams. The inhibitor does not support the slime-mould's growth, and it has different properties from previously discovered aggregation-centre inhibitors released by the amoebae themselves. The factor, as produced, does not inhibit Polysphondylium violaceum cells from forming centres or streams. Yet although E. coli itself attracts pre-aggregation P. violaceum cells, it does not mimic a P. violaceum aggregate in inducing them to form streams.

An inhibitor of cell cohesion from axenically grown cells of the slime mould, Dictyostelium discoideum.

Medium from a stationary phase culture of axenically grown D. discoideum cells contains an inhibitor of cohesion of log phase cells. The inhibitor is a heat-stable, low molecular weight substance. Its biological effects include inhibition of cohesion of aggregation-competent cells, of cells of other slime mould species, the blocking of development on Millipore filters and a reduction in adhesiv...

Cell electrophoresis of the cellular slime mould Dictyostelium discoideum. II. Relevance of the changes in cell surface charge density to cell aggregation and morphogenesis.

The reduction in the cell surface charge density of Dictyostelium discoideum during differentiation has been studied by the technique of cell electrophoresis. It was abolished under conditions in which cell aggregation was inhibited (e.g. low temperature, the presence of actinomycin D or cycloheximide). In the presence of nutrients incapable of supporting growth, cell aggregation occurred witho...

Evidence for a second chemotactic system in the cellular slime mold, Dictyostelium discoideum.

An unknown substance found in bacteria (Escherichia coli) is especially effective in attracting the vegetative amoebae of the cellular slime mold, Dictyostelium discoideum. However, the aggregating amoebae are not attracted to it at all. On the other hand, the vegetative amoebae show very little chemotactic response to cyclic adenosine monophosphate (cyclic AMP), whereas the aggregating amoebae...

A neutral ceramidase homologue from Dictyostelium discoideum exhibits an acidic pH optimum.

The nucleotide sequence reported for the Dictyostelium discoideum ceramidase is available on the DNA Data Bank of Japan (DDBJ). Ceramidases (CDases) are currently classified into three categories (acid, neutral and alkaline) based on their optimal pHs and primary structures. Here, we report the first exception to this rule. We cloned the CDase cDNA, consisting of 2142 nucleotides encoding 714 a...

A genetic study of aggregation in the cellular slime mould Dictyostelium discoideum using complementation analysis.

A series of aggregation-deficient (aggregateless) mutants were isolated in genetically marked haploid strains of the cellular slime mould Dictyostelium discoideum. Diploids were produced from pairs of such haploid mutants by a fusion system based on this organism's parasexual cycle. The diploids were isolated from the haploids by using complementation of non-allelic growth-temperature-sensitive...
